چکیده

To determine the internal structure of barley starch without amylopectin isolation, whole was hydrolyzed using ?-amylase to remove linear amylose and obtain ?-limit dextrins (?-LDs). The ?-LDs were treated with extensive ?-amylase prepare ?-limit (?-LDs), ?-LDs further into building blocks. chain-length distribution ?-LD block composition analyzed by size-exclusion chromatography anion-exchange chromatography. starches had similar pattern amylopectins conventional methods. amo1-mutated varieties contained more short B-chains less long than that other varieties. from large blocks waxy simplified method presented in this study can effectively characterize relates physicochemical properties starch, although some details are not assessable.

Plastidial Disproportionating Enzyme Participates in Starch Synthesis in Rice Endosperm by Transferring Maltooligosyl Groups from Amylose and Amylopectin to Amylopectin.

Plastidial disproportionating enzyme1 (DPE1), an α-1,4-d-glucanotransferase, has been thought to be involved in storage starch synthesis in cereal crops. However, the precise function of DPE1 remains to be established. We present here the functional identification of DPE1 in storage starch synthesis in rice (Oryza sativa) by endosperm-specific gene overexpression and suppression. DPE1 overexpre...

Enzymatic synthesis of amylopectin.

The problem of branching in glycogen and starch has been the subject of interest ever since Cori and Cori (I, 2) isolated the phosphorolytic enzyme from animal tissues and Hanes (3) found a similar enzyme in plants. Examination of polysaccharides produced by these enzymes when glucose-l-phosphate was used as a substrate showed that, similar to amylose, they had a linear configuration in which t...

Mutations in the gene encoding starch synthase II profoundly alter amylopectin structure in pea embryos.

Mutations at the rug5 (rugosus5) locus have been used to elucidate the role of the major soluble isoform of starch synthase II (SSII) in amylopectin synthesis in the developing pea embryo. The SSII gene maps to the rug5 locus, and the gene in one of three rug5 mutant lines has been shown to carry a base pair substitution that introduces a stop codon into the open reading frame. All three mutant...

A Parameterized Model of Amylopectin Synthesis Provides Key Insights into the Synthesis of Granular Starch

A core set of genes involved in starch synthesis has been defined by genetic studies, but the complexity of starch biosynthesis has frustrated attempts to elucidate the precise functional roles of the enzymes encoded. The chain-length distribution (CLD) of amylopectin in cereal endosperm is modeled here on the basis that the CLD is produced by concerted actions of three enzyme types: starch syn...
